Accuri C6 Flow Cytometer

Anonymous: This is Clare Rogers, Applications and Marketing Manager, at Accuri Cytometers. Here are some details on the C6 which hopefully address the points/concerns mentioned by Flowjock457:

1) In our white paper on the C6 design (available at our website), the patented fluidics pulse dampeners are described - invented by our chief technology officer, they function to remove the pulsation before the sheath stream arrives at the flow cell. The push-pull action of the two pumps working together creates a differential pressure which pulls sample into the flow cell; the advantage over a pressurized fluidics system is that no vacuum-seal need be formed with the sample tube (one can use any tube type that will fit and add reagents to sample tubes during data acquisition) and the volume sampled from a tube can be measured.

2) How do we get the other 3.5 logs of detection range? -The best empiric proof that the C6 has additional fluorescence-detection range due to the 24-bit DSP and low-end noise reduction that was engineered into the system, is laid out in a poster available on our website. Look under Technical Information, then Posters, titled "A Flow Cytometer with Pre-optimized Fluorescence Detectors and Broad Dynamic Range etc....". There you will see data in the far right column comparing the same sample, a mixture of unlabeled 1 micron beads and CHO cells labeled with either FITC or PE antibodies, collected on a 4-, then 5-decade instrument (at a Flow Core Facility) and then the 7-decade Accuri C6. The only instrument capable of capturing the full range of very dim fluorescence from the beads and bright fluorescence of the stained, large cell line was the Accuri C6. This shows that there is more fluorescence discrimination available on the C6 scale, even without allowing voltage adjustment, and that all decades of the scale are indeed needed and used. In fact, the point of the data in the rest of the poster is to show that setting and then monitoring the fluorescence scale, using a quality fluorescence standard, is highly beneficial for a broad array of flow applications and sample types.

(For very bright fluorescence signals, two attenuated versions of each optical filter are available (90% and 99%) - brings fluorescence signal down predictably, reproducibly by 1 or 2 logs.)

3) We have 3 US sales teams and 5 EU teams performing, on average 2-3 demos per week. Our Field Applications Specialists perform these demos by sending the C6 to the lab in a Pelican Case (via Fed Ex), unloading it onsite and being ready to run samples within an hour, max. Demos are completed in half a day. I am sure there HAVE been cases where the shipped instruments haven't performed optimally - but I can assure all that this is hardly the norm, and we would not have sold over 500 instruments world-wide if they typically arrived non-functioning to the demo site.

Anonymous: Thanks for the clarification Mr Manager, I have read a lot about this machine and it seems very good for my application. can you please state clearly how many samples does this machine process in an hour and what is the market related price. when i contacted one of your office here in a south Africa and a price of R800 000 was given to me over the phone but when i requested a quote in black and white i am quoted in the region of R1,950 000 ex vat this makes it extremely difficult for me to do the deal. thanks please contact on dixondmm@gmail
